**试验用试剂和培养基配方
 
1.磷酸盐缓冲液PBS0.03mol/LpH7.2
无水磷酸氢二钠                   2.83g
磷酸二氢钾                       1.36g
蒸馏水加至                       1000ml
将各成分加入到 1000ml 蒸馏水中,待完全溶解后,调 pH  7.27.4,于 121压力蒸气** 20min备用。
 
2.革兰染色液
    1液:结晶紫溶液
结晶紫乙醇饱和溶液                        100ml
结晶紫                                    4g8g
95%乙醇                                   100ml
1%草酸胺溶液
    2液:卢戈碘液
           碘化钾                                     2g
                                       1g
蒸馏水                                   200ml
    3液:脱色剂
195%乙醇 
2)丙酮乙醇溶液                          100ml 
95%乙醇                              70ml
丙酮                                 30ml
    4液:稀释石炭酸复红液 
碱性复红乙醇饱和溶液                     10ml 
碱性复红                                 5g10g 
5%石炭酸溶液                             90ml 
蒸馏水                                   900ml
 
3.孔雀绿与沙黄芽孢染色液
1 5.0%孔雀绿水溶液 
20.5%沙黄水溶液
 
4.无菌检验用洗脱液  
吐温-80                                   1g
蛋白胨                                    10g 
氯化钠                                    8.5g  
蒸馏水                                    1000ml
将各成分加入到 1000ml 0.03mol/L PBS液中,加热溶解后调pH 7.27.4,于121压力蒸汽**20min备用。
 
5.标准硬水(硬度342mg/L) 
氯化钙(CaCl2)                              0.034g 
氯化镁(MgCl2·6H2O)                         0.139g 
蒸馏水加至                                 1000ml
 
6.有机干扰物
        牛血清白蛋白                                  30g
        蒸馏水                                        1000ml
溶解后用微孔滤膜(孔径为0.45μm)滤过**,冰葙保存备用。
 
7.营养琼脂培养基
蛋白胨                                     10g      
牛肉膏                                     5g 
氯化钠                                     5g 
琼脂                                       15g
蒸馏水                                     1000ml
除琼脂外其他成份溶解于蒸馏水中,调 pH  7.27.4,加入琼脂,加热溶解,分装,于121压力蒸汽**20min备用。
 
8.营养肉汤培养基
蛋白胨                                     10g  
牛肉膏                                     5g 
氯化钠                                     5g 
蒸馏水                                     1000ml
将各成分溶解于蒸馏水中,调 pH 7.27.4,分装,于121压力蒸汽**20min备用。
 
9.稀释液:胰蛋白胨生理盐水溶液(TPS)。
胰蛋白胨                                         1.0g
氯化钠                                           8.5g
先用900ml以上蒸馏水溶解,并调节pH值在7.0±0.2,*终用蒸馏水加至1000ml,分装后,经121压力蒸汽**后使用。
 
10.胰蛋白胨大豆肉汤培养基(TSB
胰蛋白胨                                  1.5%
大豆蛋白胨                                0.5%
氯化钠                                    0.5%
用蒸馏水配制而成,调节pH为7.2±0.2,经121℃压力蒸汽**后使用。
 
11.胰蛋白胨大豆琼脂培养基(TSA
胰蛋白胨                                  1.5%
大豆蛋白胨                                0.5%
氯化钠                                    0.5%
琼脂                                      1.6%
用蒸馏水配制而成,调节pH为7.2±0.2,经121℃压力蒸汽**后使用。
 
12.品红亚硫酸钠培养基 
蛋白胨                                   10g 
酵母浸膏                                 5g
牛肉膏                                   5g
乳糖                                     10g
琼脂                                     15g20g
磷酸氢二钾                               3.5g
无水亚硫酸钠                             5g  
5%碱性品红乙醇溶液                       20ml 
蒸馏水                                   1000mg
    以上各成分蒸馏水溶解,调pH 7.27.4,装瓶,经121℃压力蒸汽**后使用。
 
13.溴甲酚紫蛋白胨培养液  
蛋白胨                                   10g 
葡萄糖                                   5g
可溶性淀粉                               1g
溴甲酚紫乙醇溶液                         10ml
蒸馏水                                   1000ml
将蛋白胨、葡萄糖溶解于蒸馏水中,调pH 7.07.2,加入2% 溴甲酚紫酒精溶液,摇匀后,分装(每管5ml),并放入一个小倒管,于 115压力蒸汽**30min。置4冰箱备用。
 
14.嗜热脂肪杆菌恢复琼脂培养基 
蛋白胨                                    10g
牛肉膏                                    3g
可溶性淀粉                                1g
葡萄糖                                    1g
琼脂                                      20g
蒸馏水                                    1000ml
    以上各成分蒸馏水溶解,调pH 7.07.2,装瓶,经115℃压力蒸汽**30min后使用。
 
15.需氧-***琼脂培养基
            酪胨(胰酶水解)                             15g
            牛肉膏                                     3g
            葡萄糖                                     5g
            氯化钠                                     2.5g
            L-胱氨酸                                   0.5g
            硫乙醇酸钠                                 0.5g
            酵母浸出粉                                 5g
            新鲜配制的 0.1% 刃天青溶液                 1.0ml
            (或新配制的 0.2% 亚甲蓝溶液                0.5ml
                                                  0.5g~0.7g
            蒸馏水                                     1000ml
除葡萄糖和刃天青溶液外,取上述成份加入蒸馏水中,微温溶解后,调 pH 至弱碱性,煮沸、滤清,加入葡萄糖和刃天青溶液,摇匀,调 pH 6.97.3,分装于 115℃压力蒸汽** 30min
 
16.沙堡琼脂培养基   
葡萄糖                                    40g 
蛋白胨                                    10g 
琼脂                                      20g 
蒸馏水                                    1000ml
将上述成分混合后,加热至完全溶解,调pH 5.6±0.2,于 115℃压力蒸汽** 30min备用。
 
17.沙堡液体培养基  
葡萄糖                                    40g  
蛋白胨                                    10g 
蒸馏水                                    1000ml
将上述成分混合后,加热至完全溶解,调pH 5.6±0.2,于 115℃压力蒸汽** 30min备用。
 
18.无菌试验用**培养基  
磷酸二氢钾(KH2PO4)                           1g   
硫酸镁(MgSO4·7H2O)                          0.5g 
蛋白胨                                      5g 
葡萄糖                                      10g
蒸馏水                                      1000ml
除葡萄糖外,上述各成分加入蒸馏水内,微温溶解后,调节pH 6.8,煮沸,加葡萄糖溶解后,摇匀滤清,调pH 使**后为6.4±0.2,分装,115℃压力蒸汽**20min备用。
 
19.人**细胞维持培养基  
1640干粉培养基                                                          10×10.4g
L谷氨酰胺                                                                  2.93g
丙酮酸钠                                                                     1.004g
青霉素                                                                         80万单位
链霉素                                                                         100万单位
碳酸氢钠                                                                     20.0g
Hepes                                                                           23.9g
去离子水加至                                                              10000ml
除青霉素、链霉素外,其余各成分溶于蒸馏水中,调pH7.07.2115℃压力蒸汽**20min备用。临用前加入无菌青霉素、链霉素溶液。
 
20.人**细胞完全培养基  
在人**细胞维持培养基中加入10%无菌小牛血清。
 
21.血琼脂培养基
            营养琼脂                                  100ml
            脱纤维羊血(或兔血)                         10ml
将营养琼脂加热熔化待冷至50左右,以无菌操作将10ml 脱纤维血加入后摇匀,倒平皿置冰箱备用。
 
22.霉菌培养基
                                          5g
             葡萄糖                                     10g
             磷酸二氢钾                                 1g
             氯化钠                                     5.0g
             硫酸镁                                     0.5g
             蒸馏水                                     1000ml
    除葡萄糖外,取上述成份加入蒸馏水中,微温溶解后,调 pH 6.0 ,煮沸,加入葡萄糖溶液后,摇匀,滤清,调 pH 5.66.0,分装于 115 ℃压力蒸汽**30min
 
23. SCDLP液体培养基
            酪蛋白胨                                    17g
            大豆蛋白胨                                  3g
            葡萄糖                                      2.5g
            氯化钠                                      5g
            磷酸氢二钾                                  2.5g
            卵磷脂                                      1g
            吐温80                                      7g
            蒸馏水                                      1000ml
将各种成份混合(如无酪蛋白胨和大豆蛋白胨可用日本多胨代替),加热溶解后,调pH 7.27.3,分装于121压力蒸汽**20min,摇匀,冷至 25℃使用。
 
24.伊红亚甲蓝培养基
            蛋白胨                                                                     10g
            乳糖                                                                        10g
            磷酸二氢钾                                                              2g
            2%伊红溶液                                                            2ml
            0.65%亚甲蓝溶液                                                     1ml
                                                                                  17g
            蒸馏水                                                                     1000ml
将蛋白胨、磷酸盐和琼脂溶解于蒸馏水中,调pH 7.1,分装于121压力蒸汽**20min。临用时,以无菌操作加入乳糖并加热溶化琼脂,冷至 50时,加入伊红和亚甲蓝溶液摇匀,倒平皿置4℃冰箱备用。
 
25. 0.5% 葡萄糖肉汤培养基
                                                       10g
            氯化钠                                       5g
            葡萄糖                                       5g
            肉浸液                                       1000ml
取胨与氯化钠加入肉浸液内,微温溶解后,调pH 至弱碱性,煮沸、加入葡萄糖溶解后,摇匀,滤清,调 pH 7.07.4,分装,于115压力蒸汽**30min
 
26. 甘露醇培养基
            蛋白胨                                       10g
            牛肉膏                                       5g
            氯化钠                                       5g
            甘露醇                                       10g
            0.2%溴麝香草酚蓝溶液                          12ml
            蒸馏水                                       1000ml
将蛋白胨、氯化钠、牛肉膏加入蒸馏水中,加热溶解,调 pH 7.4,加入甘露醇和溴麝香草酚蓝混匀后,分装,于 115压力蒸汽**20min
 
27. 乳糖胆盐发酵管
            蛋白胨                                       20g
            猪胆盐(或牛,羊胆盐)                           5g
                                                    10g
            0.04%溴甲酚紫水溶液                           25ml
            蒸馏水                                       1000ml
将蛋白胨、胆盐及乳糖溶解于蒸馏水中,调pH 7.4,加入0.04% 溴甲酚紫水溶液,分装(每管10ml),并放入一个小倒管,于 115压力蒸汽**15min
 
28. 乳糖发酵管
            蛋白胨                                       20g
                                                    10g
            0.04%溴甲酚紫水溶液                           25ml
            蒸馏水                                       1000ml
将蛋白胨及乳糖溶解于蒸馏水中,调pH 7.4,加入0.04% 溴甲酚紫水溶液,分装(每管10ml),并放入一个小倒管,于 115压力蒸汽**15min

苋菜红                                       1%
西曲溴胺                                     0.05%
30MEA培养基
麦芽浸膏                                     30g
大豆蛋白胨                                   3g
琼脂                                        15g
双蒸水加至                                   1000ml
将上述成份制成溶液于121℃下**15mln,**后无菌调pH5.6±0.2备用。
 
31MEB肉汤
麦芽浸膏                                      20g
双蒸水加至                                    1000ml
将上述成份制成溶液于121℃下**15mln,**后无菌调pH6.9±0.2备用。
 
32.注意事项
    (1)双料乳糖胆盐发酵管除蒸馏水外,其它成分为乳糖胆盐发酵管的2 倍;3倍浓缩乳糖胆盐发酵管除蒸馏水外,其它成分为乳糖胆盐发酵管的3倍。
    (2)配制培养基的容器不宜用铜锅或铁锅,以免影响**生长。
    (3)培养基用的试管口和三角烧瓶口应用普通棉花制成的棉塞,再用牛皮纸包好。
    (4)试剂与培养基配制好后应置清洁处保存,常温下不超过1 个月。
